<p><bold>A continuous flow synthesis of the anti-inflammatory COX-II inhibitor Celecoxib.</bold> Flow chemistry is a novel technology which can be used for the economical manufacturing of fine chemicals and active pharmaceutical ingredients (APIs) in an environment where economies of scale are not realised. Presented are our efforts to develop a rapid high-yielding flow synthesis of Celecoxib, an anti-inflammatory COX-II inhibitor.</p>

<title></title>
<p>Vloeichemie is moderne tegnologie wat gebruik kan word vir die ekonomiese vervaardiging van fyn chemikali&#x00EB;, veral vir aktiewe farmaseutiese middels in &#x2019;n omgewing waar die ekonomie van skaal gewoonlik nie gerealiseer word nie. Vloeichemie word gedefinieer as die gebruik van deurlopende prop-vloei-reaktors in plaas van konvensionele reaktors (Ley &#x0026; Baxendale <xref ref-type="bibr" rid="CIT0003">2002a</xref>; Baxendale &#x0026; Ley <xref ref-type="bibr" rid="CIT0002">2007</xref>) vir die sintese van chemiese tussenprodukte en aktiewe farmaseutiese middels.</p>
<p>Vloeichemie maak gebruik van ge&#x00EF;mmobiliseerde reagense, opruimers en vang-en-vrystel-tegnieke om van die probleme, wat tradisioneel geassosieer word met konvensionele lotsintese, soos bewerking, suiwering en optimisering, te oorkom. Tradisionele glasware word vervang deur patrone en kolomme wat voor gebruik met verskeie soliededraer-reagense gepak kan word, of deur reaksiemikroskyfies en -kronkels wat die beheerbare meng van reagense saam met uitstekende beheer van reaksietemperatuur toelaat (Baxendale et al <xref ref-type="bibr" rid="CIT0001">2006</xref>; Ley &#x0026; Baxendale <xref ref-type="bibr" rid="CIT0004">2002b</xref>).</p>
<p>Van die voordele van vloeichemie, in vergelyking met lotreaksies, lei tot ho&#x00EB;r spesifisiteit, ho&#x00EB;r opbrengs, ho&#x00EB;r deurset teen laer reaktorvolumes, laer produksiekoste, verbeterde meng- en skaalvergroting, sowel as beter gebruik van oplosmiddels en chemikalie&#x00EB;, om &#x2019;n paar te noem. Vloeichemie word wel benadeel deur duur toerusting en is slegs bruikbaar as die vorming van presipitate vermy kan word. (Ley &#x0026; Baxendale <xref ref-type="bibr" rid="CIT0003">2002a</xref>; Baxendale &#x0026; Ley <xref ref-type="bibr" rid="CIT0002">2007</xref>).</p>
<p>Ons ondersoek die gebruik van hierdie tegnologie vir die produksie van verskeie fluoorbevattende, aktiewe farmaseutiese middels met die doel om te bewys dat vloeichemie meer toepaslik as tradisionele lotsintese is. Ons pogings tot &#x2019;n vinnige, ho&#x00EB; opbrengssintese van Celecoxib, &#x2019;n anti-inflammatoriese COX-II-stremmer, sowel as die vordering met die sintese van ander fluoorbevattende, aktiewe farmaseutiese middels en tussenprodukte, word aangebied.</p>
<p>Die vloeiroete wat ontwerp is, lewer &#x2019;n ho&#x00EB; opbrengs Celecoxib, met verminderde reaksietye, en die proses word as geheel in groen oplosmiddels gedoen. Toekomstige doelwitte behels die ontwikkeling van &#x2019;n vloeiproses waardeur arielhidrasiene as arielfluoriede verkry kan word.</p>
